Let’s take a gander at some ways you can show your appreciation for the heart doctors who really make the difference in the lives of so many!The hydrolytic activity of cartilage macromolecules provides a quantitative means of evaluating tissue damage and repair in cases of trauma to the knee. The extracellular matrix of cartilage, primarily composed of cartilage proteoglycan, is thought to be the major component resisting enzymatic attack by collagenase. Since collagenase activity is believed to play a key role in the pathogenesis of joint disease, such as rheumatoid arthritis, it is important to determine the contribution of glycosaminoglycan to the inactivation of this enzyme. We have found that sulfated glycosaminoglycans significantly inhibit collagenase activity in vitro and that degradation of cartilage proteoglycan by collagenase is inhibited. However, collagenase inhibition by these glycosaminoglycans is concentration dependent and irreversible. Sulfated glycosaminoglycans will bind free collagenase, and under low ionic strength conditions, will alter the pI of the enzyme. These observations suggest that sulfated glycosaminoglycans interact directly with collagenase and represent an important means of regulating collagenase activity. The experimental system will be used to examine the kinetics and mechanism of sulfated glycosaminoglycan-collagenase interaction. We will determine whether collagenase and the
